Min-101, why is it supposed to work?

From my reading Min-101, the first drug made specifically for negative symptoms is a 5ht2a + sigma antagonist. Why is this supposed to work on negative symptoms? Every atypical antipsychotic has some degree of 5ht2a antagonism, yet they do very little for negative symptoms. Why am I supposed to get excited over and essentially tried mechanism? Someone who understands pharmacology better help me out here.

I think that research is done to understand the mechanism of the disease(schizophrenia)
@eduvigis once the mechanism is understood, and once its understood how the brain works,
true treatments can be developed it can take decades.

I just don’ understand how they are making a drug which has already been done. It just seems pointless. Like they know it will never be approved by the fda.

The sigma-2 receptor in particular is more densely located in parts of
the brain that are responsible for motor function and emotional
response.

it might work for emotional flatness …!!!

Ok, you have me interested. this sigma receptor might have some effect. but i have NEVER heard it mentioned before. are the sigma 2 antagonists we have curing all negative symptoms?

“Importantly, MIN-101 is also meant to block a specific subtype of sigma receptor called sigma2, which is involved in movement control, psychotic symptom control and learning and memory. Blocking sigma2 also modulates other neurotransmitters in the brain, in particular dopamine, which is important as individuals with schizophrenia often have elevated levels of dopamine in their brains. Blocking sigma2 also increases calcium levels in neurons in the brain, which can improve memory. Recent literature has also indicated that a sub-type of progesterone protein complex might also be a putative binding site for sigma2 receptors and might explain the effects on cognition of MIN-101.”

What about the negative symptoms? Huh? Seems like there is nothing about alogia, anhedonia, avolition etc

We need something for cognition in schizophrenia!
Something that reverses brain damage!
@eduvigis
and is free of side effects.

haha sz can’t reverse…!!! but the mystery will soon become reveled to us …!!! dont worry…!!

u confused me edu …!!! U might have a greater understanding then me…!!! how is ur semester going on …!! I cant study due to sz…!!!

What understand far cry is that this sigma 2 receptor is good for learning and memory but it says nothing about negative symptoms. Learning and memory are not negative symptoms.

Edit: My semester is up and down, thanks for asking. Sometimes I have negative symptoms (caused by medication) but for a period of time during the day they are gone. Then I study. I am just doing online classes, not real in person classes. I don’t have the social skills for that.

yes its so much mysterious…!!!

If they don’t know how to treat negative symptoms why do they even bother?

I am also confused as to how Min-101 is supposed to work, but so far clinical trials have proven its efficacy for negative symptoms, which is enough for me.

yes it is due to complexity of brain and less people suffer from SZ only1%…!!! scientist lack fund for research …i am hoping board institute will break another mystery on sz soon ,!! keep hoping …!!!
